چکیده
The problem of determining the molecular weight of DNA samples by sedimentation equilibrium in a buoyantcdensity gradient is considered for the case of DNA samples with density heterogeneity. By determining apparent molecular weights in two or more buoyant mediums, quantitative measure of the amount of density heterogeneity can be determined. This method may be employed to determine both the true molecular weight and the extent of base composition heterogeneity.

The approach to equilibrium in a buoyant-density gradient.
The approach to equilibrium in a buoyant-density gradient is a transport process in a centrifugal field and may be used to measure a sedimentation coefficient. This process is interesting for several reasons. Densitygradient sedimentation equilibrium can be used for the determination of the molecular weight of high-molecular weight DNA samples homogeneous in buoyant density. Furthermore, buoyan...
متن کاملMolecular weights of homogeneous coliphage DNA's from density-gradient sedimentation equilibrium.
The problem of obtaining meaningful molecular weights for high molecular weight DNA samples by sedimentation equilibrium in a density gradient has been solved by correcting the theory for thermodynamio non-ideality. Several different methods of virial corrections are suggested and examined to determine the best method of obtaining molecular weights at infinite dilution. A pronounced virial effe...
متن کامل[ 6 ] Density Gradient Sedimentation Equilibrium
In 1957, Meselson, Stahl, and Vinograd 1 introduced the remarkable technique of sedimentation equilibriuln in a density gradient for the study of the properties of high molecular weight DNA in solution. Because of the very high molecular weights of DNA, conventional sedimentation equilibrium cannot be used; for even at the lowest centrifugal fields which are practical, the force on these molecu...
متن کاملSedimentation Equilibrium in a Density Gradient
Sedimentation equilibrium in a density gradient has recently been developed into a reliable method for the determination of molecular weights of homogeneous DNA samples. The procedure and calibrations required for this method are outlined in this paper. In 1957, Meselson, Stahl and Vinograd' introduced a remarkable analytical technique for studying the properties of DNA and viruses. Sedimentati...
متن کاملMethylmercury-induced sedimentation heterogeneity of T7 bacteriophage DNA.
Single-stranded and methylmercurated T7 DNA is composed of two species as evidenced from the sedimentation pattern displayed during band-sedimentation in self-generating density gradients as well as during equilibrium-sedimentation in CS2SO4 density gradients. Under the given experimental conditions, viz., at pH 9.18 and in presence of 0.1 mM CH3HgOH, the two species band in CS2SO4 with a densi...
